Induced resistance to 6-methylpurine and cycloheximide in Tetrahymena. II. Variation within vegetative cultures of micronucleate T. thermophila and amicronucleate T. pyriformis.

چکیده

Resistance to 6-methylpurine and cycloheximide has been induced in both the micronucleate species Tetrahymena thermophila and the amincronucleate species T. pyriformis. Resistance follows only after mutagen treatment and vegetative growth. The frequencies with which resistant variants are induced and the independence of mutagenesis and selection are demonstrated. All evidence is consistent with the hypothesis that macronuclear subunits are assorting in both species during vegetative growth to produce new phenotypes among subclones.

The vegetative micronucleus has a critical role in maintenance of cortical structure in Tetrahymena thermophila.

The vegetative role of the germinal micronucleus of Tetrahymena thermophila was studied. Amicronucleate cells that had spontaneously arisen in star strains (very aged cones with defective micronuclei) were observed in B* as well as in A* and in C* and were found to lack the oral apparatus and to have disordered ciliary rows. To reduce uncertainty given the very small micronucleus and possible e...

Effect of different toxic compounds on ATP content and acid phosphatase activity in axenic cultures of Tetrahymena pyriformis.

The sensitivity of protozoa, and particularly ciliated protozoa, to environmental changes suggested a study on the physiological responses arising from exposure to toxic compounds. Tetrahymena pyriformis was used as a test organism in a set of miniaturized assays. The physiological response of this ciliate was assessed in terms of adenosine-5'-triphosphate content and acid phosphatase activity ...

A NEW BIOLOGICAL SCREENING SYSTEM FOR LOCAL ANAESTHETICS BY INHIBITION MOBILITY OF TETRAHYMENA PYRIFORMIS

An alternative in vitro approach to drug screening has been the use of human cell cultures for antiviral agents and microbial cell cultures for the assessment of the carcinogenic potential of selected compounds. A number of protozoan species have been also used as drug screens for anti-protozoal agents. The ciliated protozoan Tetrahymena pyriformis species has been widely utilised as a dru...

Cloning and characterization of the major histone H2A genes completes the cloning and sequencing of known histone genes of Tetrahymena thermophila.

A truncated cDNA clone encoding Tetrahymena thermophila histone H2A2 was isolated using synthetic degenerate oligonucleotide probes derived from H2A protein sequences of Tetrahymena pyriformis. The cDNA clone was used as a homologous probe to isolate a truncated genomic clone encoding H2A1. The remaining regions of the genes for H2A1 (HTA1) and H2A2 (HTA2) were then isolated using inverse PCR o...

Structure of Macronuclear Envelopes of Tetrahymena Pyriformis in the Stationary Phase of Growth

In a preceding publication a method was reported by which fragments of the nuclear envelopes of Tetrahymena pyriformis could be isolated (10). When macronuclear envelopes were isolated from exponentially growing cultures of the amicronucleate Tetrahymena pyriformis strain GL, two types of envelope pieces were observed which differed markedly in the structure of their pore complexes. Type A cont...
